Yes, focus peaking works in manual focus mode. You are right about it, but the reason I use 49-area is that 80% chance it works(by touch) like 1-area and 20% it helps lock on somewhere near to start with, just personal experience. All it does is attempt to focus on 49 areas and select one (or if they're at the same distance from the camera, more than one) to focus on quite randomly. These works fine for me, but with one down side(or a bug, and I believe it's a shutter AF OFF bug), the focus peaking won't engage, so there won't be highlight(peaking) around focused subject. MF overrides the Shutter AF setting, so Manual focus=back button, Auto=Traditional-half-press for shutter and exposure, full to shoot. If you switch (as I do) between back button and traditional, you can leave Shutter AF at ON.
